摘要: Tooth enamel has the highest degree of biomineralization all vertebrate hard tissues. During secretory stage formation, ameloblasts deposit an extracellular matrix that is in direct contact with ameloblast plasma membrane. Although it known integrins mediate cell–matrix adhesion and regulate cell signaling most types, receptors production are not well characterized. We hypothesized αvβ6 integrin expressed where regulates enamel. Human mouse were found to express both β6 mRNA protein. The maxillary incisors Itgb6−/− mice lacked yellow pigment their mandibular appeared chalky rounded. Molars showed signs reduced mineralization severe attrition. mineral-to-protein ratio was significantly enamel, mimicking hypomineralized amelogenesis imperfecta. Interestingly, amelogenin-rich abnormally accumulated between layer forming surface, also ameloblasts. This accumulation related increased synthesis amelogenin, rather than removal proteins. confirmed cultured ameloblast-like cells, which endocytosis receptor for amelogenins, although participated on this indirectly via endogenously produced In summary, by plays a crucial role regulating amelogenin deposition and/or turnover subsequent biomineralization.
