The proteome profile of two cell lines and their xenografts isolated from a patient with clear cell sarcoma (soft tissue melanoma).

摘要: We report the establishment of two novel clear cell sarcoma (CCS) lines (soft tissue melanoma) from a patient and production corresponding xenografts after xenotransplantation those cells to NOD/SCID mice. As no comprehensive study on relevant proteomes this type cancer has been reported date, proteomics technologies were applied in first attempt analyze proteins their primary xenografts. Total protein extracts separated by dimensional gel electrophoresis (2-DE) analysed MALDI-MS MALDI-MS-MS following in-gel digestion with trypsin. Protein identification was carried out peptide mass fingerprint (PMF) post source decay (PSD), respectively. Comparative analysis revealed that 124 common between xenografts; 249 found be expressed only proteome lines, while 178 Our results demonstrated both positive for vimentin S100 as markers CCS. After functional analysis, 27 different groups identified proteomes, including apoptosis-related proteins, oncogenes several closely related TP-53 NF-kappa B pathways. Furthermore, nestin, stem growth factor CLC11 mdr-1, malignant-melanoma-initiating cells, Since there are data concerning expression CCS, may contribute understanding molecular basis disease, well developed used tools development new therapeutic strategies tackle rare but fatal malignancy.
