The cognitive psychopharmacology of Alzheimer's disease: focus on cholinergic systems.

摘要: The primary pathology in Alzheimer's disease (DAT) occurs the basal forebrain cholinergic system (BFCS), which provides major innervation to neocortex, hippocampus and amygdala. Consistent with 'cholinergic hypothesis' of dementia DAT, most effective treatments so far developed for DAT are drugs act boost functions BFCS. These include centrally acting cholinesterase inhibitor tacrine, agonist nicotine, acute administration leads an improvement attentional functions, line recent animal studies role BFCS cognition. We conclude that future research should development more potent, longer-lasting, less toxic agents, appear be best candidates alleviating cognitive symptomatology DAT. Such may also useful treatment a number other disorders, including Lewy body dementia, attention deficit/hyperactivity disorder, schizophrenia.
