摘要: Abstract By means of new or modified procedures troponin-tropomyosin complex was fractionated into three components plus tropomyosin, and then reconstituted in the absence urea. Tropomyosin separated from troponin by hydroxyapatite column chromatography, a method giving sharper separation than previously used technique isoelectric precipitation. The chromatography also tropomyosin two fractions. One gave single band on sodium dodecyl sulfate gel electrophoresis, other double band; complete proline suggested that each different form pure tropomyosin. Troponins I, T, C were DEAE-Sephadex 6 m urea as previously, but found to be much improved if treated with Ca2+ chelator before being applied column. activity fractions assayed at varying ratios actin both without present. It near physiologic actin, either combination I T inhibited acto-heavy meromyosin ATPase. However, cases when C, which is necessary for sensitivity, added, inhibition reversed not only presence Ca2+. Only present addition did occur We therefore conclude first, may required second, all are restore full third, this reconstitution can accomplished combining individual
