摘要: CD8+ T lymphocytes play a critical role in immunity to Trypanosoma cruzi. However, the target molecules of this cell subset have not been elucidated. In work, we report identification an H-2Kb-restricted CTL epitope within two trypomastigote surface Ags encoded by members T. cruzi sialidase/trans-sialidase gene superfamily. Octapeptide VDYNFTIV sensitized cells for lysis generated from spleens cruzi-infected mice. Peptide-specific lines were cytotoxic, secreted IFN-gamma and TNF-alpha, but low undetectable levels IL-4 IL-5, able, upon adoptive transfer, confer high degree protection against challenge infection. Finally, protective determinant appears be conserved among parasites diverse geographic locations. This constitutes first identified class I MHC-restricted provides basis search additional targets considered development vaccines Chagas' disease.
