The cGMP-gated Channel and Related Glutamic Acid-rich Proteins Interact with Peripherin-2 at the Rim Region of Rod Photoreceptor Disc Membranes

摘要: The rod cGMP-gated channel is localized in the plasma membrane of photoreceptor outer segments, where it plays a central role phototransduction. It consists alpha- and beta-subunits that assemble into heterotetrameric protein. Each subunit contains structural features characteristic nucleotide-gated channels, including cGMP-binding domain, multiple membrane-spanning pore region. In addition, beta-subunit has large glutamic acid- proline-rich region called GARP also expressed as two soluble protein variants. Using monoclonal antibodies conjunction with immunoprecipitation, cross-linking, electrophoretic techniques, we show associates Na/Ca-K exchanger segment membrane. This complex proteins interact peripherin-2 oligomers rim disc membranes. These results suggest channel/peripherin interactions mediated by part play connecting discs to anchoring channel.exchanger
