Modified rapid immunohistochemical staining for intraoperative diagnosis of malignant brain tumors.

摘要: Rapid immunohistochemistry (R-IHC) has been developing mainly as a support technique in the rapid diagnosis of central nervous system tumors; however, there have problems regarding instability specimen preparation and immunostaining. To overcome weakness this technology, immunostaining, we developed modified R-IHC. This was achieved by switching to 4% paraformaldehyde fixative solution utilizing commercially available Polymer Refine Detection Kit, high-sensitivity kit, place secondary antibodies. In study, tested R-IHC evaluating immunostaining on new staining items 94 brain tumor removal cases, which took at Tokyo Women's Medical University from 2014 2015. The results showed that, based GFAP p53 markers, method obtained higher stability specimens than standard method. It also stainability same level that permanent specimen. 86.6% (46/53) 82.8% (24/29) pHH3 ATRX, respectively, percentage correct classification (PCC) against specimens, 100% (7/7) PCC malignant lymphomas gliomas used CD20/CD3 for discrimination. We concluded indicated comparison GFAP, p53, CD20/CD3, pHH3, ATRX.
