摘要: The c-erbB-2 gene has been found amplified in a number of human adenocarcinomas leading to elevated levels expression the p185 protein product. Increased this putative growth factor receptor reported occur by molecular mechanism other than amplification and for reason we have studied normal colon lesions representing different stages neoplastic progression. We report 3 44 carcinomas 1 5 preneoplastic polyps studied. Confirmation product was established Western blot analysis immunocytochemical staining tissue sections. An extended study, involving adenomatous carcinomatous material immunostaining, revealed detectable presence 20% carcinomas, consistent with immunoprecipitation data derived using cell lines. In contrast, high percentage showed strong both antibodies used, indicating associated lesions. Staining restricted localization cells on luminal colonic surface, no crypt. Histologically mucosa, adjacent tumor, more extensive distribution crypt suggestive disturbance c-erbB-2. These results indicate that is early neoplasia but do not establish it as primary these events. occurrence multiple copies lesions, however, suggests possible role some malignancies.
