Expression of Nuclear Receptors and Apo E Secretion During the Differentiation of Monocytic THP-1 cells into Macrophages

摘要: The human monocytic THP-1 cell line differentiates into macrophage-like cells that secrete apo E after addition of PMA. Using this model, we studied the time course transcriptional activation and secretion in relation with expression nuclear receptors. Upon treatment PMA, mRNA protein were triggered concomitant increase LXRalpha, PPARgamma, PPARbeta levels. PPARalpha was downregulated, RXRalpha unchanged, RARalpha VDR showed only transient increases. FXR SXR transcripts not detectable. Specific agonists used to investigate functional role these receptors upon secretion. LXRalpha ligands T0901317 22(R)-hydroxycholesterol most potent inducers, followed by PPARgamma agonist BRL49653.
