ALTERED PLASMA ZINC LEVEL CONTRIBUTES TO THE DEVELOPMENTAL TOXICITY OF VALPROIC ACID IN SKELETAL SYSTEM OF RAT

摘要: Valproic acid is one of the main antiepileptic drugs. There an increased risk neural tube defects and axial skeletal malformations among infants born to women who had received valproic acid. There a hypothesis that biochemical abnormality underlying teratogenicity a drug-induced reduction in maternal plasma zinc .In present experimental study mated rats were divided into four groups 8 animals each [control, (VPA), + (VPA+ Zn) and (Zn) groups]. The VPA group 300 mg/kg acid, daily.The control group received equal volume 0.9% NaCl. VPA+ Zn 30 mg/kg zinc sulfate sulfate, daily. NaCl, were administered intraperitonealy from day 6 through 15 gestation. On 16, six group were authanized other were scarified on gestational (GD) 20 evaluate skeletal system elder fetuses. Blood was drawn determine zinc. data analyzed by using analysis variance (Kruskal -Wallis test). concentration treated with valproic significantly lower than those 16 GD (P=0.004). Some anomalies such as hydrocephaly, spina bifida, hemivertebrate, rib seen VPA treated group. Low percentage anomalies bifida observed treated group while no groups. results present experiment support abnormalities causing of VPA drug–induced deficiency, possibly, it may also result of embryonic Zn.
