Review Article Beyond laser microdissection technology: follow the yellow brick road for cancer research

摘要: Normal biological tissues harbour different populations of cells with intricate spacial distribution pat- terns resulting in heterogeneity their overall cellular composition. Laser microdissection involving direct viewing and expertise by a pathologist, enables access to defined cell or specific region on any type tissue sample, thus selecting near-pure targeted cells. It opens the way for molecular methods directed towards well-defined populations, provides also powerful tool studies focused limited number has wide applications oncology (diagnosis research), bio- logy, biochemistry forensics selection, but other areas have been gradually opened up these new methodological approaches, such as cultures cytogenetics. In clinical trials, profiling microdissected samples can yield global "omics" information which, together, morphological analysis cells, provide basis diagnosis, prognosis patient-tailored treatments. This remarkable technology brought insights understanding DNA, RNA, functions regulation proteins identify disease signatures. We review herein laser variety fields, we particularly focus attention pre-analytical steps that are crucial successfully per - form molecular-level investigations.
