摘要: Abstract To investigate the transcriptional mechanisms of rice tungro bacilliform virus, we have systematically analyzed an activator element located immediately upstream TATA box in virus promoter and its cognatetrans-acting factors. Using electrophoretic mobility shift assays, showed that nuclear proteins bind to element, forming multiple specific DNA-protein complexes via protein-protein interactions. Copper-phenanthroline footprinting DNA methylation interference analysis indicated share a common binding site between positions −60 −39, contact major groove. UV cross-linking assays further two (36 33 kDa), found cell suspension shoot extracts, one (27 present root this element. In protoplasts derived from (Oryza sativa) culture, is prerequisite for activity function critically dependent on position relative box. Thus, activation may interactions with basal machinery, propose mediated by position-dependent mechanism.
