摘要: Summary Heterologous genes introduced into the nuclear genome of Chlamydomonas reinhardtii are often poorly expressed. To understand molecular mechanisms underlying this effect, we examined influence various factors on expression a chimeric transgene that confers resistance to zeomycin. This marker comprises bacterial ble gene flanked by 5′ and 3′ sequences from RBCS2 gene. We found frequency with which transformants recovered is significantly increased when fused shorter versions promoter introns coding region ble. The latter effect particularly evident in case first intron RBCS2, dramatically stimulates transformation level expression. improvement mediated part an enhancer element within sequence, acts orientation-independent manner effective placed either upstream or downstream promoter. Our results demonstrate stable high-level foreign possible, highlight potential role as modulators alga.
