摘要: Abstract Anandamide ( N -arachidonoylethanolamine, AEA) is an endocannabinoid present in human plasma that associated with several physiological functions and disease states. Significant variability AEA concentrations has been reported between studies, because quantification of fraught methodological difficulties. A rapid, highly sensitive, robust, specific, reproducible ultra high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method described here for the analysis plasma. This fully validated using octa-deuterated (AEA-d8) as internal standard represents improvement over previous analyses terms run time (4 min), limit detection (0.055 fmol on column, 18.75 fmol/ml plasma), precision (relative deviations 3.7, 3.9, 4.8% 1.66, 6.65, 133 fmol column), accuracy (97.5–104.5%). was linear range 0.23 to 19 nM (1.66 column). To demonstrate usefulness this measurement levels clinical samples, samples obtained from female volunteers at different stages menstrual cycle pregnant women were assayed. Plasma significantly P  = 0.0078) lower luteal phase compared follicular phase. In pregnancy, lowest first second trimesters comparable those observed modestly increased third trimester. The highest active labour, these  = 0.0147) higher than term but not labour. Postmenopausal had during premenopausal  = 0.0389) sensitivity suggests suitable may be utilised investigation biomatrices containing limited amounts AEA.
