摘要: The cDNA for the developmentally regulated, neurite outgrowth-promoting protein HB-GAM (heparin-binding growth-associated molecule) was recently cloned and shown to encode a novel lysine-rich sequence that is homologous with retinoic acid-induced sequences suggested function in cell differentiation (Merenmies, J., Rauvala, H. (1990) J. Biol. Chem. 265, 16721-16724). same found mitogenic pleiotrophin (Li, Y.-S., Milner, P. G., Chauhan, A. K., Watson, M. A., Hoffman, R. M., Kodner, C. Milbrandt, Deuel, T. F. Science 250, 1690-1694). In this study, we have constructed recombinant baculovirus using encodes putative preprotein of HB-GAM. secretion signal cleaved off expression system, rapidly secreted culture medium. Recombinant purified from medium retains biochemical characteristics activity tissue-derived protein. Studies on suggest functions as an extracellular matrix-associated enhances axonal growth perinatal cerebral neurons rat. Since predicted amino acid has been ascribed protein, activities fractions were also studied. did not display any significant activity, suggesting preparations may contain other heparin-binding factors. We identified brain-derived 17-kDa (p17) detached heparin by slightly higher salt concentration compared p17 structurally distinct responsible actions fractions.
