摘要: A procedure is described for intensifying histochemical reactions by amplification of biotinylated sites. This achieved deposition tyramine on the tissue through enzymatic action horseradish peroxidase (HRP). The amplified biotin sites are subsequently visualized binding them to avidin, which a marker attached. greatly increases sensitivity staining procedures that employ HRP (and/or biotin) in tissue. For neuroanatomical pathway tracing methods, detectability injected tracer. lectin histochemistry and immunohistochemistry, requires or primary antibody be diluted. dilution results less background yet strong signals produced even when very dilute reagents used. Alternatively, permits much shorter incubations antibodies dilutions used would ordinarily with conventional bridge techniques. also useful amplifying weak signals, such as those immunoreactions glutaraldehyde-fixed procedure, together availability avidin probes labeled fluorochromes, colloidal gold, enzyme systems other than HRP, provides means increasing versatility variety reactions, including detecting situ hybridization probes, addition reactions.
