摘要: Due to constraints in vector construction, reporter polypeptides often carry N-terminal sequences of extraneous origin. Since protein half-life can be influenced by small determinants the N-terminus, such foreign destabilize proteins and compromise results reporter-based studies. We provide a real-life example this problem (destabilizing derived from ribosomal protein) show that it solved with ubiquitin fusion technique, which are placed upstream reporter, our case beta-glucuronidase. Post-translational processing characterized pathways removes together destabilizing sequences, generating stable whose N-terminus is constant.
