Distribution of the class II beta-tubulin in developmental and adult rat tissues.

摘要: During a screen of monoclonal antibodies raised against cytoskeletal preparation neonatal rat cerebrum, we have identified antibody, MAb58A, that is specific for the class II beta-tubulin isotype. Immunoscreening brain cDNA library using MAb58A yielded retaining II-specific nucleotide sequence. The specificity to isotype was confirmed by immunoreactivity synthetic peptides corresponding isotype-specific sequence I, II, III, IVa, or IVb. Further, results an immunoassay series overlapping octapeptides derived from region revealed antibody epitope heptapeptide consists Glu-Glu-Glu-Glu-Gly-Glu-Asp (EEEEGED). Immunoblot analysis represented significant portion present in adrenal gland, brain, and testis adult rats. In fetal tissues, this detected skeletal muscle, as well brain. Immunohistochemically, reacted predominantly with components developing nervous system, such migrating neuroblasts, peripheral nerves ganglion cells, sensory organs. MAb58A-immunoreactivity also found smooth muscle chondrocytes, vascular endothelia. adults, remarkably diminished, but persisted capillary components. Together, our demonstrate isotype, which may retain embryonic nature both neuronal non-neuronal tissues.
