Dynamic interactions between L-type voltage-sensitive calcium channel Cav1.2 subunits and ahnak in osteoblastic cells

摘要: Voltage-sensitive Ca2+ channels (VSCCs) mediate permeability in osteoblasts. Association between VSCC α1- and β-subunits targets channel complexes to the plasma membrane modulates function. In mechanosensitive tissues, a 700-kDa ahnak protein anchors VSCCs actin cytoskeleton via β2-subunit of L-type Cav1.2 (α1C) complex. is major α1-subunit osteoblasts, but cytoskeletal complex subunit composition are unknown. Among four β-subtypes, and, lesser extent, β3-subunit coimmunoprecipitated with MC3T3-E1 preosteoblasts. Fluorescence resonance energy transfer revealed β2-subunits demonstrated their association secretory pathway. Western blot immunohistochemistry showed β2-subunit. Cytochalasin D exposure disrupted did not disassemble or disrupt function ahnak. Similarly, small interfering RNA knockdown significantly impaired influx. Collectively, we that form stable osteoblastic cells permits signaling independently cytoskeleton.
