摘要: Purpose Myopia, or nearsightedness, is characterized by excessive lengthening of the ocular globe and associated with extracellular matrix remodeling in posterior sclera. The activity gelatinase A, a member metalloproteinase family, has been shown to increase sclera during development induced myopia several species. In present study, distribution relative expression A its inhibitor, tissue inhibitor metalloproteinases (TIMP)-2, were measured within fibrous scleras experimentally myopic (form-deprived) eyes, control eyes recovering from form deprivation better understand mechanisms that regulate scleral rate elongation. Methods Total RNA was extracted form-deprived chick myopia, paired contralateral subjected northern blot analysis analyses using cDNA probes chicken TIMP-2. TIMP-2 mRNAs evaluated situ hybridization on frozen sections 33P-labeled probes. Gelatinase gelatin zymography. Results Northern indicated increased 128% deprived (P = 0.009), whereas after 1 day recovery, levels decreased 80% 0.005). contrast, significantly (-53%, P 0.027) eyes. No significant differences detected between most transcripts layer Conclusions Changes steady state mRNA lead changes mediate, at least part, process visually regulated growth remodeling.
