Alterations in classical cadherins associated with progression in ulcerative and Crohn's colitis

摘要: Human colitis is a condition associated with spectrum of altered morphologic changes and cellular adhesion. The role cadherins, which are powerful morphoregulatory cell adhesion molecules, in provocative as yet unknown. Herein, we present results that suggest strong correlation between the deregulation two cadherin E- P-cadherins, progression human colitis. We examined expression structural integrity P-cadherins inflamed, dysplastic, or neoplastic ulcerative (UC) (n=58), Crohn's (n = 30), normal tissue 20) to assess function abnormal epithelium. E-cadherin strongly expressed colorectal epithelium, whereas left-sided UC it either down-regulated has single-base pair mutation exon 4 resulting an amino acid alteration (6 58 cases). By contrast, P-cadherin dramatically up-regulated both disease especially dysplastic tissue. In vitro transfected SW-480 cells containing mutations identical those vivo were increased spontaneous disaggregation compared wild-type E-cadherin. Based on this evidence, hypothesize small subset expressing mutant widespread ulceration, rapidly dividing immature phenotype includes dysplasia. differential mutated cadherins herein broad epithelial phenotypes, lymphocyte integrin binding, resistance denudation, seen adenocarcinoma sequence.
