摘要: The aggregation and deposition of amyloid-β (Aβ) peptides are believed to be central events in the pathogenesis Alzheimer’s disease (AD). Inoculation brain homogenates containing Aβ aggregates into susceptible transgenic mice accelerated deposition, suggesting that capable self-propagation hence might prions. Recently, we demonstrated can monitored live using bioluminescence imaging (BLI). Here, use BLI probe ability self-propagate following inoculation bigenic mice. We report compelling evidence prions by demonstrating widespread cerebral β-amyloidosis induced either purified derived from or composed synthetic Aβ. Although were sufficient induce vivo, they exhibited lower specific biological activity compared with brain-derived aggregates. Our results create an experimental paradigm should lead identification self-propagating conformations, which could represent novel targets for interrupting spread AD patients.
