Pulmonary response to inhaled antigen: neuroimmune interactions promote the recruitment of dendritic cells to the lung and the cellular immune response to inhaled antigen.

摘要: Dendritic cells (DCs) play a critical role in capturing and presenting inhaled antigens to T lymphocytes. We report that pulmonary DCs the Lewis rat are normally located lung immediate proximity nerve fibers contain immunoreactive substance P (SP). Functionally, bound 125I-SP displayed increased motility vitro response graded concentrations of SP. However, SP had no effect on accessory cell activities DCs. To examine neural influences immune antigen, rats were pretreated with capsaicin (CAP), which damages small nerves depletes neuropeptide stores, then challenged intratracheally (i.t.) hen egg lysozyme (HEL). The number antigen-presenting CAP-treated comparable those controls up day 14. lymphocytes harvested from regional lymph nodes draining effectively sensitized HEL both groups. when i.t. at 0 rechallenged 14, lungs showed decreased numbers OX-6+ diminished lymphoid infiltrates compared controls. suggest CAP interferes neural-mediated contributes accumulation inflammatory during efferent limb pulmonary-cell-mediated vivo.
