摘要: Substantial mortality of in vitro manipulated porcine embryos is observed during peri-attachment development. Herein we describe our efforts to characterize the transcriptomes embryonic disc (ED) and trophectoderm (TE) cells from derived vivo fertilization, fertilization (IVF), parthenogenetic oocyte activation (PA), somatic cell nuclear transfer (SCNT) on days 10, 12, 14 gestation. The IVF, PA, SCNT were generated with matured oocytes cultured overnight before being transferred recipient females. Sequencing cDNA resulting samples was accomplished Genome Analyzer IIx platform Illumina. Reads aligned a custom-built swine transcriptome. A generalized linear model fit for ED TE separately, accounting embryo type, gestation day, their interaction. Those genes significant differences between types characterized terms gene ontologies KEGG pathways. Transforming growth factor-β signaling downregulated EDs IVF embryos. In embryos, ubiquitin-mediated proteolysis ErbB aberrantly regulated. Expression involved chromatin modification, silencing by RNA, apoptosis significantly disrupted summary, have used high-throughput sequencing technologies compare expression profiles various We expect that these data will provide important insight into root causes (and possible opportunities mitigation of) suboptimal development assisted reproductive technologies.
