Heat Shock Alters Keratocyte Movement and Morphology: Exploring a Role for HSP27 (HSPB1)

摘要: HSP27 is essential for mammalian cell movement. To further explore the effects of heat shock and mechanistic role HSP27, we have initiated a study using well-established model rapidly moving cells, fish keratocyte. Here report that causes decrease in speed. Since changes morphology can drastically affect movement, also monitored morphology. Heat caused number polar cells an increase those with one stuck adherent edge, indicating occurrence both cytoskeletal re-organization increased adhesion to substrata. Analyses levels Western blots showed they were relatively high keratocytes prior remained afterward. In contrast, blot analysis HSP70 it was induced strongly by shock, mounted robust response. Surprisingly, given propensity localize nuclear/perinuclear regions following location unchanged as shown indirect immunostaining anti-HSP27 antibodies. Fluorescence intensities immunostained images before after quantified Image J software. The results this fluorescence intensity decreased suggesting affected antibody recognition. Possible roles regulating actin filament dynamics, speed are discussed.
