摘要: Background: A potent angiogenesis inhibitor U-995, has been purified from the cartilage of blue shark (Prionace glauca). U-995 is composed two single peptides with molecular mass 10 and 14 kDa, respectively. Materials Methods: was designed to study human umbilical vein endothelial cell (HUVEC) migration proliferation in vitro induced by TNFα chicken chorioallantoic membrane (CAM). Furthermore, we determined ability inhibiting tumor growth metastasis. Results: (15 30 μg/ml) markedly inhibited HUVEC and, at 15-50 μg/ml produced a dose-dependent decline [ 3 H]-thymidine incorporation. 50 when added TNFα-induced caused discontinuous disrupted blood vessels. Moreover (30 prevented collagenase-induced collagenolysis. In addition, 200μg injected i.p. into mice it suppressed sarcoma-180 B16-F10 mouse melanoma metastasis vivo. Conclusions: These results suggest that antiangiogenic effects may be due interference HUVECs as well inhibition collagenolysis, thereby leading both growth.
