摘要: Background Major depressive disorder is a worldwide neuropsychiatric associated with various symptoms, but current antidepressants used in clinical practice have side effects and high failure rates. Andrographolide the main bioactive ingredient of Andrographis paniculata exhibits numerous pharmacological actions. This study aimed to evaluate antidepressant-like andrographolide male C57BL/6J mice. Methods The mice were explored forced swim test, tail suspension chronic unpredictable mild stress model depression. Western blotting immunofluorescence further performed assess on brain-derived neurotrophic factor signalling cascade hippocampal neurogenesis. Moreover, inhibitor (K252a) lentiviral-short hairpin RNA (LV-TrkB-shRNA) clarify mechanism andrographolide. Results exhibited potential test without influencing locomotor activity Repeated treatment not only produced significant also prevented decreasing neurogenesis Importantly, blockade system by K252a TrkB-shRNA fully abolished Conclusions exerts via promoting cascade.
