Doxorubicin plus Interleukin-2 Chemoimmunotherapy against Breast Cancer in Mice

摘要: As recently characterized, following s.c. implantation into syngeneic C57BL/6 mice, E0771 tumor invades locally dermal layers and peritoneum, metastasizes to the lung, induces a nonspecific immunosuppression in host. Using this breast medullar adenocarcinoma model, therapy consisting of single moderate dose doxorubicin followed by twice daily doses interleukin-2 for 30 days was examined efficacy mechanism action when given animals with established disease. This combination treatment, but not combinants alone, resulted tumor-free long-term survival 40% mice without significant toxicity 83% survivors had immune memory specific cells. Treatment also decreased suppression induced tumor. Full response treatment required functional CD8+ T cells, whereas depletion natural killer cells caused only reduction rate. A serum “biomarker” profile that correlated with, seemed predictive of, identified nuclear magnetic resonance–based metabonomic analysis.
