Diferential adriamycin® istribution to blood components

摘要: The differential distribution of doxorubicin (Adriamycin® = AM) and daunorubicin (Daunomycin DM) within the blood components, after an i.v. injection 10 or 15 mg/kg body weight, was investigated in Cd-COBS rats. AM DM were measured by a fluorimetric procedure, unchanged compound separated from its metabolites quantified means TLC scanning fluorescence technique. accumulated following order (of decreasing percentages): plasma red cells (RBC), white (WBC), platelets (PT), but absolute amount drug that reached each cell type related to relative volume. In presence higher concentrations (after weight) RBCs much more than plasma, WBC, PT; suggesting RBC fraction has greater capacity concentrate drug. However, if concentration is expressed per unit volume component, markedly value observed for PT, this confirmed byin vitro results obtained incubating AM. seemed be distributed on percent basis extent fraction. Both compounds taken up cells, particularly platelets, levels excess extracellular concentration.
