摘要: Abstract Objective The aim of this study was to assess the potential store-operated Ca 2+ channel (SOC) antagonists as purging agents for leukemia cells. Materials and Methods Clonogenic, limiting dilution, nuclear condensation assays were used evaluate SOC antagonist efficacy. activity endoplasmic reticulum content measured by flow cytometry. Murine bone marrow transplantation determine efficacy effects on hemopoietic reconstitution. Results Econazole (Ec) ketotifen (Ke) variably effective against human murine cell lines after 24 hours incubation. However, a 2-hour serum bovine albumin-free treatment protocol with Ec found maximize differential sensitivity between leukemic cells normal progenitors. Primary acute myelogenous blast viability reduced 4.2 5.1 logs dilution. An inverse relationship Ke in untransformed observed. Nuclear condensation, an index apoptosis, which occurred 24-hour treatments either or Ke, not observed serum- exposures; however, condensed nuclei additional 10-hour incubation growth medium without drug. Using deliberately contaminated 1% P815 cells, we showed that highly vitro can be accomplished using no adverse reconstitution mice. Conclusion These studies suggest have residual present context high-dose chemotherapy autologous leukemia.
