摘要: Abstract Background The BK polyomavirus (BKPyV) is subdivided into four genotypes. consequences of each genotype and donor-recipient (mis)match for BKPyV-associated nephropathy (BKPyVAN) in kidney transplant recipients (KTRs) are unknown. Objectives To develop evaluate a genotype-specific IgG antibody-based BKPyV serotyping assay, order to classify donors accordingly. Study design VP1 antigens six variants (Ib1, Ib2, Ic, II, III IV) were expressed as recombinant glutathione-s-transferase-fusion proteins coupled fluorescent Luminex beads. Sera from 87 healthy blood 39 KTRs used analyze seroreactivity serospecificity against the different Six sera with marked serotype profiles analyzed further pseudovirus neutralizing capacity. Results Seroreactivity was observed all genotypes, seropositivity rates above 77% comparable donors. Strong cross-reactivity (r > 0.8) among I subtypes, genotypes IV. Seroresponses IV seemed genuine, while those II could be out(cross)competed. GMT (Luminex) IC50 (neutralization assay) values showed good agreement determining strongest seroresponse within an individual. Conclusions Despite some degree cross-reactivity, this assay seems useful tool identify main infecting given This information, which cannot obtained otherwise nonviremic/nonviruric individuals, provide valuable information regarding prevalent warrants study.
