摘要: The Notch receptor initiates a unique intercellular signaling pathway that is evolutionarily conserved across all metazoans and contributes to the development maintenance of numerous tissues. Consequently, many diseases result from aberrant signaling. Emerging roles for in disease are being uncovered as studies reveal new information regarding various components this pathway. activity regulated at several levels, but O-linked glycosylation Epidermal Growth Factor (EGF) repeats extracellular domain has emerged major regulator that, depending on context, can increase or decrease activity. Three types occur consensus sequences found within EGF Notch: O-fucosylation, O-glucosylation, O-GlcNAcylation. Recent have investigated site occupancy these also defined specific glycans structure function. Nevertheless, there functional aspects each type O-glycosylation remain unclear. Here, we will discuss molecular mechanisms how regulates describe disorders associated with defects O-glycosylation.
