摘要: We have isolated and characterized Mpp10p, a novel protein component of the U3 small nucleolar ribonucleoprotein (snoRNP) from yeast Saccharomyces cerevisiae. The MPP10 was first identified in human cells by its reactivity with an antibody that recognizes specific sites mitotic phosphorylation. To study functional role pre-rRNA processing, we performing GenBank search. Mpp10p homolog is 30% identical to over length. Antibodies purified recognize 110-kDa polypeptide extracts immunoprecipitate snoRNA, indicating snoRNP yeast. As step genetic analysis function, diploid S. cerevisiae were transformed null allele. Sporulation tetrad indicate essential gene. A strain constructed where expressed galactose-inducible, glucose- repressible promoter. After depletion growth glucose, cell arrested levels 18S 20S precursor are reduced or absent while 23S 35S precursors accumulate. This pattern accumulation rRNA suggests required for cleavage at A0, A1, A2. Pulse-chase newly synthesized pre-rRNAs Mpp10p-depleted confirms little mature formed. These results reveal ribosome biogenesis further elucidate composition snoRNP.
