Prevalence of deletions of YY1‐binding sites in episomal HPV 16 DNA from cervical cancers

摘要: Expression of the oncogenes E6 and E7 human papillomavirus 16 (HPV 16) appears enhanced in pre-malignant malignant genital tumors. We recently identified a transcriptional silencer upstream oncogene promoter P97, comprising 4 binding sites for cellular YY1 protein. The analysis long control regions (LCR) episomal HPV DNAs from primary tumors lymph-node metastases 6 patients with cervical cancer revealed deletions point mutations cases. To test activity P97 promoter, mutated LCRs were cloned luciferase reporter gene vector. A mutation YY1-recognition site 4, which prevents DNA-protein interaction, did not affect activity, probably due to compensation by overlapping YY1-binding 3. However, 5.5- 6.5-fold increased expression was obtained under 3 shortened lacking 2 sites. 2, previously shown stimulate 3.5-fold, could be detected both tumor metastasis, indicating that is stable characteristic DNA associated individual cancer. These findings suggest or play significant role over-expression viral progression.
