摘要: Publisher Summary This chapter presents the quantification and structural elucidation of low quantities radiolabeled metabolites using microplate scintillation counting (MSC) techniques in conjunction with liquid chromatography-mass spectrometry (LC-MS). Radioactive isotopes, mainly 14C, are routinely used to trace fate drugs disposition metabolism studies conducted during their development. Information derived from these is a critical component developing safe effective new also regulatory requirement for registration approval. MSC an off-line chromatographic detector that has been available only past few years; however, short time it shown great promise as powerful versatile tool detection quantitative analysis levels metabolites. Compared conventional HPLC-LSC methods, HPLC-MSC not comparable precision, better sensitivity increased analytical throughput, significantly reduces radioactive waste requires minimal manual operation. coupled electrospray ionization mass spectrometer ideal choice abundance Additionally, LC-MSC high mass-resolution MS-based defect filtering technique selective metabolite ions complex biological matrices.
