摘要: Objective To investigate whether treatment with anti-vascular endothelial growth factor (VEGF)-neutralizing antibodies can reduce pain and voiding dysfunction in the cyclophosphamide (CYP) cystitis model of bladder mice. Materials Methods Adult female mice received anti-VEGF-neutralizing (10 mg/kg i.p. B20-4.1.1 VEGF mAb) or saline (control) pre-treatment, followed by CYP (150 mg/kg i.p.) to induce acute cystitis. Pelvic nociceptive responses were assessed applying von Frey filaments pelvic area. Spontaneous micturition was using void spot assay. Results Systemic antibody significantly reduced response compared control (saline). In anti-VEGF pre-treatment group, there a significant increase hypersensitivity, measured area under curve (AUC) at 5 h post-CYP administration (P = 0.004); however, 48 h 96 h administration, hypersensitivity had 54% 47%, respectively, time point, no longer different from baseline 0.22 0.17, respectively). There difference urinary frequency mean voided volume between two groups. Conclusion Systemic blockade signalling effective reducing pelvic/bladder pain.
