摘要: Secretory leukocyte peptidase inhibitor (SLPI) plays a role in proliferation and differentiation via the autocrine paracrine systems. SLPI's expression is well-documented reproductive tract, but it remains unclear whether active during early embryonic development. In this study, of Slpi embryo were evaluated. vitro cultures chemically defined simple medium resulted reduction developmental speed from 8-cell stage, as well implantation rate compared with vivo embryos. SLPI protein was localized to membrane or submembrane cytoplasm an stage-dependent manner. cultured embryos exhibited lower levels mRNA than knockdown by antisense oligonucleotides attenuated sense oligonucleotide-transfected controls. The critical period for attenuation occurred after stage. treatment accelerated development, increased rate, ameliorated suppressive effects knockdown. did not induce changes total cell number inner blastocysts. Meanwhile, upregulated factors matrix metalloproteinase-14, neutrophil elastase, tissue metalloproteinase-1. Together, these results suggest that effective regulator competence manner, respectively, controlling development factors, such MMP family members.
