摘要: The CFA/I fimbria promotes the attachment of enterotoxigenic Escherichia coli (ETEC) to surface human enterocytes. generation a protective immune response requires induction antibodies able block CFA/I-mediated binding ETEC receptors located on small intestine epithelium or red blood cells, in hemagglutination tests. An eukaryotic expression plasmid, pBLCFA, encoding gene under control cytomegalovirus major immediate-early promoter was constructed as prototype DNA vaccine against ETEC. pBLCFA-tranfected BHK-21 cells secreted peptide cross-reacting with monoclonal antibody raised subunits. BALB/c mice immunized intramuscularly one two doses purified pBLCFA developed CFA/I-specific serum for at least 52 weeks, composed predominantly IgG1 subclass. pBLCFA-induced bind mainly epitopes exposed intact fimbriae and do not react recessive found other fimbra sharing amino acid homologies CFA/I. Furthermore, were adhesive properties fimbriae, evaluated by ability inhibit promoted CFA/I-expressing cells.
