摘要: Cell therapy is a rapidly growing area of research for the treatment osteoarthritis (OA). This work aimed to investigate efficacy intra-articular adipose-derived stromal cell (ASC) injection in healing process on cartilage, synovial membrane and menisci an experimental rabbit model. The induction OA was performed surgically through bilateral anterior cruciate ligament transection (ACLT) achieve eight weeks from ACLT mild grade OA. A total 2 × 106 6 autologous ASCs isolated inguinal fat, expanded vitro suspended 4% serum albumin (RSA) were delivered hind limbs; RSA used as control. Local bio-distribution cells verified by injecting chloro-methyl-benzamido-1,1'-dioctadecyl-3,3,3'3'-tetra-methyl-indo-carbocyanine per-chlorate (CM-Dil) labeled limbs. Cartilage histological sections scored Laverty's scoring system assess severity pathology. Protein expression some extracellular matrix molecules (collagen I II), catabolic (metalloproteinase-1 -3) inflammatory (tumor necrosis factor- α) markers detected immunohistochemistry. Assessments carried out at 16 24 weeks. Labeled-ASCs unexpectedly medial meniscus but not cartilage tissue 3 20 days ASC-treatment. Intra-articular ASC administration decreases progression exerts contribution treated animals comparison groups. Our data reveal capacity promoting repair attenuating events inhibiting progression. On basis local findings, benefits obtained could be due trophic mechanism action release growth factors cytokines.
