RNA interference revealed the roles of two carboxylesterase genes in insecticide detoxification in Locusta migratoria.

摘要: Carboxylesterases (CarEs) play key roles in metabolism of specific hormones and detoxification dietary environmental xenobiotics insects. We sequenced characterized CarE cDNAs putatively derived from two different genes named LmCesA1 LmCesA2 the migratory locust, Locusta migratoria, one most important agricultural pests world. The full-length (1892 bp) (1643 encode 543 501 amino acid residues, respectively. deduced CarEs share a characteristic α/β-hydrolase structure, including catalytic triad composed Ser-Glu (Asp)-His consensus sequence GQSAG, which suggests that both are biologically active. Phylogenetic analysis grouped into clade A has been suggested to be involved detoxification. Both transcripts were highly expressed all nymphal adult stages, but only slightly eggs. Analyses tissue-dependent expression situ hybridization revealed primarily gastric caeca. RNA interference (RNAi) followed by topical application carbaryl or deltamethrin did not lead significantly increased mortality with either insecticide. However, RNAi insect mortalities 20.9% 14.5%, respectively, when chlorpyrifos was applied. These results suggest these might significant role likely L. migratoria.
