摘要: The electroconvulsive threshold (ECT) test is used commonly in the screening of anti-epileptic drugs rodent models, but little known about its genetic or mechanistic basis. Thresholds for minimal clonic, maximal tonic, psychomotor (partial) seizures were determined 16 different inbred mouse strains two laboratories. A wide range thresholds was observed, suggesting that a variety neuroexcitability alleles exist strains. Although there generally good cross-strain correlation between three seizure types, several outlier detected, showing genetically encoded differences can affect ability particular type to spread through brain. Furthermore, relative susceptibility strain comparable laboratories, despite sites, instrumentation, and personnel, ECT assay portable common often be relied upon as calibration standards. Last, paradigm also sensitive enough detect single locus differences, laying groundwork mutation screens new models.
