摘要: The Staphylococcus aureus MSCRAMM (microbial surface components recognizing adhesive matrix molecules) protein clumping factor A (ClfA) has been shown to be a critical virulence in several experimental models of infection. This report describes the generation, characterization, and vivo evaluation murine monoclonal antibody (MAb) against ClfA. Flow cytometric analysis revealed that MAb 12-9 recognized ClfA expressed by all clinical S. strains obtained from variety sources. In assays measuring whole-cell binding human fibrinogen, inhibited over 90% displaced up 35% previously adherent bacteria. Furthermore, single infusion was protective an intravenous challenge with methicillin-resistant strain sepsis model (P < 0.0001). These data suggest anti-ClfA should further investigated as novel immunotherapy for treatment prevention life-threatening infections.
