摘要: An expression cloning screen was used to isolate a novel gene homologous the extracellular cysteine-rich domain of frizzled receptors. The (which we called sizzled for secreted frizzled) shown encode soluble protein, containing functional signal sequence but no transmembrane domains. Sizzled (szl) is capable inhibiting Xwnt8 as assayed by (1) dose-dependent inhibition siamois induction in animal caps, (2) rescue embryos ventralized DNA and (3) XmyoD marginal zone. Szl can dorsalize Xenopus if expressed after midblastula transition, strengthening idea that zygotic wnts particular plays role antagonizing dorsal signals. It also suggests ventralizing parallels opposition BMPs noggin chordin. szl restricted narrow ventral zone gastrulating embryos. thus encodes antagonist wnt signaling likely involved ventrally whose represents new element molecular pattern
