摘要: Raman optical activity (ROA) is an advanced technique capable of detecting structural deformations light-absorbing molecules embedded in chromophoric proteins. Resonance (RR) spectroscopy widely used to enhance the band intensities. However, theoretical work has predicted that under resonance conditions ROA spectrum resembles shape RR spectrum. Herein, we use photoactive yellow protein (PYP) measure first experimental data on effect changing excitation wavelength spectra a protein. We observe close similarity between and PYP. Furthermore, experimentally verify prediction concerning ratio amplitudes spectra. Our demonstrate selecting appropriate key factor for extracting information active site using spectroscopy.
