摘要: The use of an on-line adsorption preconcentrator coupled with capillary zone electrophoresis/nanoelectrospray mass spectrometry (PC/CZE/nESMS) is described for the analysis peptides and protein digests. investigation was focused on production disposable preconcentrators made large particle size (40 microns irregular packing), thereby eliminating a retaining frit without loss performance. These preconcentration devices were commercially available components which can be easily interfaced to current CZE/nESMS systems. Practical issues such as composition stationary phase, elution volume sample breakthrough carry-over evaluated in order optimize analytical performance this technique. Under optimized conditions, PC/CZE/nESMS technique provided separation efficiencies excess 100,000 theoretical plates loading 8 microliters. Sample minimized by proper reconditioning prior CZE separation. Alternatively, resulting from small volumes could used advantageously provide multiple analyses single injection sample. application demonstrated proteolytic Bauhinia purpurea lectin at concentration level 30 nM. Further structural information obtained using tandem elucidate structure N-linked glycans amino acid sequences glycopeptides.
