摘要: We have developed and evaluated a real-time reverse transcriptase PCR (RT-PCR) assay for the detection of human enterovirus D68 (EV-D68) in clinical specimens. This was response to unprecedented 2014 nationwide EV-D68 outbreak United States associated with severe respiratory illness. As part our evaluation outbreak, we sequenced published genome sequence virus circulating St. Louis, MO. sequence, along other GenBank sequences from past occurrences, used computationally select region appropriate targeting strain-specific RT-PCR assay. The amplifies segment VP1 gene, an analytic limit 4 copies per reaction, it more sensitive than commercially available assays that detect enteroviruses rhinoviruses without distinguishing between two, including three multiplex panels approved use by FDA. did not any or tested divergent strains EV-D68, first strain (Fermon) identified California 1962. should be useful identifying studying current future outbreaks viruses.
