Pro-Apoptotic Activity of 4-Isopropyl-2-(1-Phenylethyl) Aniline Isolated from Cordyceps bassiana

摘要: Cordyceps species including bassiana are a notable anti-cancer dietary supplement. Previously, we identified several compounds with activity from the butanol fraction (Cb-BF) of bassiana. To expand structural value Cb-BF-derived drugs, employed various chemical moieties to produce novel derivative, KTH-13-amine-monophenyl [4-isopropyl-2-(1-phenylethyl) aniline (KTH-13-AMP)], which tested for activity. KTH-13-AMP suppressed proliferation MDA-MB-231, HeLa, and C6 glioma cells. also dose-dependently induced morphological changes in cells time-dependently increased level early apoptotic stained annexin V-FITC. Furthermore, levels active full-length forms caspase-3 caspase-9 were increased. In contrast, total caspases-3, caspase-8, caspase-9, Bcl-2 decreased treated-cells. We confirmed that phosphorylation STAT3, Src, PI3K/p85, is linked cell survival, was diminished by treatment KTH-13-AMP. Therefore, these results strongly suggest this compound can be used guide development an drug or serve as lead forming another strong anti-proliferative agent.
