摘要: Purpose Raman spectroscopy is a promising cancer detection technique for surgical guidance applications. It can provide quantitative information relating to global tissue properties associated with structural, metabolic, immunological and genetic biochemical phenomena in terms of molecular species including amino acids, lipids, proteins, nucleic acid (DNA). To date vivo Raman systems mostly included probes biopsy needles typically limited single-point interrogation over scale between 100 500 microns. The development wider field handheld could improve tumor localization range open surgery applications brain, ovarian skin cancers. Methods Here we present novel implementation using coherent imaging bundle fibers create probe capable reconstructing images mesoscopic fields view. Detection performed linear scanning rotation mirror an spectrometer. Different slits widths were tested at the entrance spectrometer optimize spatial spectral resolution while preserving sufficient signal noise ratios detect principal features. non-biological samples Calcite Polytetrafluoroethylene (PTFE) used characterize performance system. new wide-field was on ex calf brain swine tissue. content both types validated data from literature compared acquired probe. as gold standard against which instrument benchmarked since it has already been thoroughly biological characterization. Result We have developed characterized practical non-contact imager providing 115 microns view >14 mm2 6 cm−1 whole fingerprint region. Typical integration time acquire entire image set approximately s. Spectra (single point wide-field) showed good agreement, Pearson correlation factor > 0.85 different categories. Protein lipid imaged manifested into measured spectra correlated well previous findings literature. An example map presented based ratio protein-to lipid-content showing clear delineations white gray matter adipose muscle tissue.
