摘要: Biochemical and morphological evidence is presented to support the characterization of a rat bone-marrow-derived cell line (RPM) as an analog promegakaryoblast. The conditions for in vitro growth maturation RPM are described. Rapid proliferation readily achieved when cultures supplemented with moderate levels fetal bovine serum (FBS). proliferative compartment small blast-like cell. Immunofluorescent staining demonstrates that cells contain factor VIII:antigen fibrinogen their cytoplasm. secrete, into conditioned medium, potent mitogenic activity aortic smooth muscle cells. When incubated under relative deprivation, stop proliferating undergo process maturation. sequence described stage I (promegakaryoblast--the compartment); II (immature megakaryocyte or promegakaryocyte); III (mature megakaryocyte). release, from cytoplasm, membrane-bound vesicular bodies containing lavender granules cytoplasmic organelles. These have been designated IV. may provide useful model study synthesis megakaryocyte-specific proteins.
