摘要: Sample preparation is key to the success of proteomics studies. In present study, two sample methods were tested for their suitability on mature, recalcitrant leaves six representative perennial plants (grape, plum, pear, peach, orange, and ramie). An improved method was obtained: Tris Triton X-100 added together instead CHAPS lysis buffer, a 20% TCA-water solution 100% precooled acetone after protein extraction further purification protein. This effectively eliminates nonprotein impurities obtains clear two-dimensional gel electrophoresis array. The facilitates separation high-molecular-weight proteins increases resolution low-abundance proteins. provides widely applicable economically feasible technology proteomic study plants.
